Genetic polymorphism data on 15 autosomal STR markers in a Western Romanian population sample

Abstract

The allelic frequency distribution and statistical genetic parameters of forensic relevance for 15 STR loci (D8S1179, D21S11, D7S820, CSF1PO, D3S1358, TH01, D13S317, D16S539, Penta E, Penta D, vWA, TPOX, D18S51, D5S818 and FGA) in a population sample of 336 non-related individuals residing in the Western part of Romania are presented.

    No difference was observed with Turkey 1, Turkey 2, Iraq, and Azerbaijan population in the 15 STR loci. Additionally comparisons of FST genetic distance were observed for Eastern Turkey with other populations by means of published allele frequency data (Bulbul et al., 2014; Cakir et al., 2003; Gurkan et al., 2015; Barni et al., 2007; Hedjazi et al., 2013; Sánchez-Diz et al., 2008; Nasibov et al., 2013; Ghada et al., 2009; Anghel et al., 2014; Brisighelli et al., 2009; Borosky et al., 2014; Ozeki and Tamaki, 2013) (Table 4). The following significant differences (p < 0.05) are found: 1 out of 15 loci in Iran (D16S539), 1 out of 15 loci in Azerbaijan (D8S1179), 2 out of 15 loci in Turkısh Cypriots (D16S539, FGA), 2 out of 15 loci in Greece (D13S317, FGA), 3 out of 15 loci in Iraq (D3S1358, D13S317, D19S433), 3 out of 15 loci in Egypt (D21S11, THO1, D5S818), 3 out of 13 loci in Romanian (D8S1179, THO1, D13S317), 3 out of 13 loci in Italy (D8S1179, FGA, D5S818), 7 out of 15 loci in Argentina (D21S11, D7S820, THO1, D13S317, D19S433, D5S818, FGA) and 13 out of 15 loci in Japanese (D21S11, D7S820, CSF1PO, D3S1358, THO1, D13S317, D16S539, D2S1338, D19S433, vWA, TPOX, D18S51, D5S818).

    We first investigated the frequencies of 15 autosomal STR loci and the results are shown in Fig. 1A and Supp. Table S1. In accordance with the previous reports of STR locus frequency, pairwise GD (genetic distance) values between the Chinese group and 8 neighboring groups (Romanian, Estonian, Iranian, Cayman Islander, South African, Brazilian, Libyan and Korean) were calculated and the data is shown in Supp. Table S2 (Lee et al., 2010; Aguiar et al., 2012; Khodjet-el-Khil et al., 2012; Hedjazi et al., 2013; Anghel et al., 2014; Lucassen et al., 2014; Sadam et al., 2014; Faris and Tanzillo-Swarts, 2015). Phylogenetic reconstruction was conducted to illustrate the genetic relationships and the UPGMA tree is shown in Fig. 1B. From our results, the shortest distance between the Chinese group and the other groups was 0.0073 (compared with Korean groups).

    Nearest affinity can be seen between the Guangdong populations and the Yunnan population. The population pairwise genetic distance based on the allele frequencies was calculated using published population data including Japanese [14], Koreans [15], Philippine [16], African American [17], Caucasian [17], Hispanic [17], Western Romanian [18], German [19] and Maghreb [20] (Table S2). When compared with the Japanese, Caucasian and Western Romania population, the allele frequency distributions of the Guangdong Han population showed significant differences at the all same detected loci.
